Serine/threonine-protein kinase 3 (UniProt: Q13188; also known as EC:2.7.11.1, Mammalian STE20-like protein kinase 2, MST-2, STE20-like kinase MST2, Serine/threonine-protein kinase Krs-1) is encoded by the STK3 (also known as KRS1, MST2) gene (Gene ID: 6788) in human. STK3 is a homodimeric protein that serves as a stress-activated, pro-apoptotic kinase. It is expressed at high levels in adult kidney, skeletal and placenta tissues and at very low levels in adult heart, lung, and brain tissues. Full activation of STK3 requires its homodimerization and autophosphorylation at threonine 180. Caspase-3 dependent cleavage of STK3 produces a 36 kDa (MST-2/N) and a 20 kDa ST2/C form. Following its cleavage by caspase-3 between residues 322-323, the 36 kDa (MST-2/N) form enters the nucleus and induces chromatin condensation followed by internucleosomal DNA fragmentation. Its phosphorylation at threonine 117 causes inhibition of its cleavage, kinase activity, nuclear translocation, and autophosphorylation at threonine 180. STK3 is a key component of the Hippo signaling pathway that plays a pivotal role in organ size control and tumor suppression by restricting cell proliferation and promoting apoptosis. The activated STK3 kinase negatively regulate the yes-associated protein 1 (YAP1) and WW Domain-containing transcription regulator protein 1 (TAZ), which trigger the expression of proliferation genes and inhibit apoptosis to promote carcinogenesis. However, it is also reported to promote the migration of CD8+ cells by activating NF-kB signaling and suppress ovarian tumor growth. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
