General description

ヒト線維芽細胞・上皮細胞共培養系由来のコラーゲンIVは、in vitro ECM（細胞外マトリックス）を形成し、3次元細胞培養の優れた生物学的スキャフォールドとして機能します。

コラーゲンIVは、2本のα1様鎖と1本のα2様鎖を含むヘテロ三量体分子です。胚形成の完了に必要であると考えられており、適切な組織構造および構造的完全性にとって必要です。 in vitroにおいて、多くの種類の細胞の付着と増殖を強化する基質として使用されます。ヒト線維芽細胞および上皮細胞が産生したコラーゲンIVは、さらに生化学的に精製され、25%酢酸中で濃度0.3 mg/mlの滅菌溶液として提供されます。

COL4A1（コラーゲンタイプIV α1鎖）は、コラーゲンファミリーに属しています。N-末端の7Sドメイン、C-末端の球状ドメイン（NC1）、およびGly-X-Yリピートの短い挿入をともなう中心の三重らせん部分を持っています[1]。基底膜におけるタイプIVコラーゲンの重要な成分です。COL4A1遺伝子は、ヒトクロモソーム13q34に位置します。

Application

ヒト培養細胞由来コラーゲンタイプIVは、iPSC（人工多能性幹細胞）の培養および分化に使用されています。また、膜のコーティングにも使用されています。[1]

Biochem/physiol Actions

COL4A1（コラーゲンタイプIV α1鎖）関連の疾患は、肺合併症に関連しています。この遺伝子は、基底膜の安定性に必要な主成分です。COL4A1における変異は、家族性脳孔症、小児片麻痺、脳小血管病（CSVD）、および出血性卒中を引き起こします。

Features and Benefits

このIV型コラーゲン製品は、in vitroでECMを産出する共培養系のヒト線維芽細胞および上皮細胞から生成されます。
次に、IV型コラーゲンを生化学的に精製します。
酢酸および0.5%クロロホルム溶液中で透析により滅菌され、25%酢酸中で濃度~0.3 mg/mlの滅菌溶液として提供されます。
