Gene Information

human ... EIF4EBP1(1978)

General description

Eukaryotic translation initiation factor 4E binding protein 1 (E1F4EBP1) is part of the translation repressor protein family. The gene encoding it is localized on human chromosome 8p11-p12.

Biochem/physiol Actions

Eukaryotic translation initiation factor 4E binding protein 1 (E1F4EBP1) is involved in protein synthesis, cell growth and survival. In the unphosphorylated form, it suppresses the translation initiation of capped mRNAs, whereas in phosphorylated form, it increases translation initiation. Phosphorylation of E1F4EBP1 releases it from eukaryotic translation initiation factor 4E (eIF4E) and this results in cap-dependent translation. The protein is also involved in tumorigenesis and the gene is overexpressed in various carcinomas. It is also a substrate for mTORC1 (mammalian target of rapamycin complex 1). In non-phosphorylated conditions, E1F4EBP1 interacts with p21 (cyclin-dependent kinase inhibitor) and enhances destabilization of p21.

Two related eukaryotic initiation factor-4E binding proteins (4E-BP1 and 4E-BP2) were recently characterized for their capacity to bind specifically to eIF4E and inhibit its function. Here, we determined the cDNA sequence, tissue distribution, genomic structure, and chromosome localization of murine
