General description

Tissue inhibitor of metalloproteinase-2 (TIMP-2) is an extracellular inhibitor of matrix metalloproteinases (MMPs) including MMP1, 2, 3, 7, 8, 9, 10, 12, 13, 14, 15, 16 and 19. It belongs to the I35 (TIMP) family of irreversible protease inhibitors. The gene encoding this protein is localized on human chromosome 17q25. Recombinant human TIMP-2 is a 21.8kDa protein containing 194 amino acid residues.

Biochem/physiol Actions

Tissue inhibitor of metalloproteinase-2 (TIMP-2), like other protease inhibitors, functions as a key modulator of extracellular matrix degradation during tissue development and remodeling. TIMP-2 can also act through a matrix metalloproteinase (MMP)-independent mechanism inhibiting endothelial cell proliferation in vitro. It demonstrates anti-angiogenic activities in vivo. TIMP-2 is a target gene of the microRNA-miR-22.

Physical form

Lyophilized from 10 mM Sodium Phosphate, pH 7.5.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
