General description

Human Jak3 (GenBank Accession No. NM_000215), (amino acids 781-end) with N-terminal His tag, MW = 37 kDa, expressed in Baculovirus expression system.

Application

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.

Unit Definition

One unit is defined as the amount of enzyme that will phosphorylate 1 nmol of tyrosine substrate per minute at pH 7.4 and 30°C.

Physical form

Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 20% glycerol and 1 mM DTT.

Preparation Note

Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Janus kinases (JAKs) are nonreceptor protein tyrosine kinases that play a role in a broad range of cell signaling. JAK2 and JAK3 have been involved in the pathogenesis of common lymphoid-derived diseases and leukemia cancer. Thus, inhibition of both JAK2
