EN219 is a cell-permeable chloroacetamide compound that selectively inhibits E3 ligase RNF114-mediated autoubiquitination and p21 ubiquitination. EN219 is shown to covalently interact with RNF114-C8 (IC50 = 470 nM), TUBB1-C201, HSPD1-C442, and HIST1H3A-C97. EN219 is a recruiter of the E3 ubiquitin ligase RNF114 that can be utilized for PROTAC applications.
