Biochem/physiol Actions

NCTT-956 is a very potent, specific inhibitor of 12-lipoxygenase (12-LO) activity, with an IC50 of 800 nM, no activity against 5-LO and greater than 25-fold selectivity over 15-LO. Specific inhibition of 12-LO by NCTT-956 in platelets blocks PKC activation, aggregation, granule secretion and αIIbB3 activation.

Oxidative stress and inflammation are important factors contributing to the pathophysiology of numerous neurological disorders, including Alzheimer's disease, Parkinson's disease, acute stroke, and infections of the brain. There is well-established evidence that proinflammatory cytokines and glutamate, as well as reactive
