General description

Ammonium pyrrolidinedithiocarbamate is a nuclear factor κB (NF-κB) inhibitor.[1] It functions as a chelating agent, which allows the preconcentration of trace metals from solution.[2] Pyrrolidinedithiocarbamate (PDTC) has antioxidant properties. It inhibits apoptosis in lymphocytes, neurons and vascular endothelial cells.[3]

Application

Ammonium pyrrolidinedithiocarbamate has been used:
  • to inhibit nuclear factor κB (NF-κB) mobilization and TNF (tumor necrosis factor) production in human monocytes[4]
  • to reduce neutrophil activity in rats[5]
  • to prevent Ca2+ influx in adriamycin (ADR)-treated podocytes[6]

to abrogate the microRNA-146a promoter luciferase reporter activity in human brain cells[7]

Biochem/physiol Actions

Prevents induction of nitric oxide synthetase by inhibiting translation of NOS mRNA; induces apoptosis in rat smooth muscle cells and inhibits apoptosis in leukemia HL-60 cells.

  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.

    1. Sigma-Aldrich has not performed a controlled solution stability test on PDTC. However, we have found that aqueous stock solutions are typically stable for at least one month when stored at -20 °C.

    1. PDTC has been reported to repress vascular cell adhesion molecule-1 (VCAM-1) activation by Interleukin-1β in cultured human umbilical vein endothelial calls. Marui, M., et al., J. Clin. Invest, 92, 1866 (1993).  It inhibits the induction of nitric oxide synthase activity in rat alveolar macrophages. Sherman, M.P., et al., Biochem. Biophys. Res. Commun. 191, 1301-1308 (1993).  It is an Inhibitor of nuclear factor kappa B., Eberhardt, et al. Biochem. Biophys. Res. Commun. 200, 163 (1994).
