Phosphoglucose Isomerase ubiquitous multifunctional cytosolic protein. This dimeric protein has a molecular weight of approximately 119,500 ± 600 and is composed of four subunits that are nearly identical.

Application

Phosphoglucose Isomerase from baker′s yeast (S. cerevisiae) has been used:
  • to convert fructose-6-phosphate to glucose-6-phosphate during the enzymatic measurement of serum mannose levels.
  • as one of the enzymes in the series of four enzyme-coupled reactions to measure phosphomannomutase (PMM) activity.
  • to convert fructose to glucose while evaluating fructose content in kudzu roots.

Biochem/physiol Actions

Phosphoglucose Isomerase(PGI) is an enzyme crucial for the interconversion of D-glucose 6-phosphate and D-fructose 6-phosphate. PGI is responsible for the second step of glycolysis and is involved in glucogenesis and pentose phosphate pathway (PPP). It is highly conserved in bacteria and eukaryotes. PGI plays a vital role in the survival of A. fumigatus and demonstrates diverse functions that encompass development, biosynthesis of cell wall glucan, and virulence.Phosphoglucose Isomerase functions as an isomerase, neuroleukin, autocrine motility factor, and a differentiation and maturation mediator.

Unit Definition

One unit will convert 1.0 μmole of D-fructose 6-phosphate to D-glucose 6-phosphate per min at pH 7.4 at 25 °C.
