General description

Calsequestrin (CASQ) 1 gene is mapped to human chromosome 1q21, a region associated with type 2 diabetes in Amish and other population. The gene codes for a calcium storage protein, CASQ1, expressed in sarcoplasmic reticulum of fast-twitch skeletal muscle cells.

Application

Biochem/physiol Actions

Calsequestrin (CASQ) 1 is one of the major components of the protein backbone of luminal Ca2+ reservoir. It plays a crucial role in calcium flux by regulating calcium channel activity and associating with other proteins localized in the sarcoplasmic reticulum. CASQ1 is involved in insulin- and/or contraction-stimulated glucose transport. Mutation of this gene leads to the accumulation of CASQ1 and other proteins in sarcoplasmic reticulum (SR) vacuole, which results in vacuolar myopathy. Experimental studies on mice shows that CASQ1 is involved in the maintenance of calcium homeostasis and size of terminal cisternae (TC) in skeletal muscle fibers. Alteration in the expression of CASQ1 affects GLUT4 recruitment and insulin sensitivity. Single-nucleotide polymorphisms in the calsequestrin-1 gene act as a genetic marker for graves′ ophthalmopathy (GO) and hashimoto′s thyroiditis (HT).
