General description

The gene cerebellar degeneration-related protein-2 (CDR2) has been mapped to human chromosome 16p12.3. The gene encodes a cytoplasmic protein harboring a helix-leucine zipper motif. CDR2 is localized in the cytoplasm. The mRNA is ubiquitously expressed in adult mouse tissue with most abundance in the testis. However, the protein is present predominantly in brain, testis, gynecological and breast tumors, suggesting post-transcriptional regulation. CDR2 is cell cycle regulated in tumor cells with highest expression in mitosis.

Application

Biochem/physiol Actions

Cerebellar-degeneration-related antigen-2 (CDR2) is a target antigen in paraneoplastic cerebellar degeneration (PCD). In PCD, CDR2 has been shown to activate specific cytotoxic T lymphocytes which are likely to contribute to the development of autoimmune neuronal degeneration. CDR2 is up-regulated in breast and ovarian tumor cells obtained from the general population of cancer patients. In addition, it is shown to attenuate hypoxia response in renal cell carcinoma by regulating prolyl-4-hydroxylase domain protein abundance. Through its leucine zipper motif CDR2 interacts with c-myc in adult cerebellar Purkinje neurons, tumor cell lines and cerebellar extracts. CDR2 down-regulates c-myc-dependent transcription. The protein is required for mitosis, as CDR2 knockdown cells have aberrant mitotic spindles and impaired proliferation.
