General description

TAF4 (TATA-box binding protein associated factor 4) is a component of transcription factor complex TFIID, which forms an essential part of the transcriptional machinery. TFIID contains TATA-binding protein (TBP) and 14 TAFs. It contains two copies of TAF4. The N-terminal of TAF4 contains a conserved glutamine rich domain. Its core is composed of co-activator TAF4-homology ETO-TAFH/CRI/NHR1 (TAFH) domain, and its C-terminal contains a histone-like (CRII) domain.

Application

Biochem/physiol Actions

TAF4 (TATA-box binding protein associated factor 4) is responsible for maintaining the structural stability of the TFIID complex. It mediates interactions with CREB (cAMP response element-binding) and Sp1 (specificity protein) proteins, and as it is a part of the Wnt signaling, it plays a role in cell fate determination and differentiation and development. Its controlled degradation is linked to proper male germ-cell differentiation, and TAF4b is essential for the development of oocytes and spermatogenesis. It interacts with activation function-2 (AF-2), and induces the transcription of retinoic acid, vitamin D3 and thyroid hormone receptors. It is also involved in the regulation of neurongeneis and neuronal differentiation, where it interacts with TP53 and noncanonical Wnt signaling.
