General description

78 kDa glucose-regulated protein (UniProt: P11021; also known as GRP-78, Endoplasmic reticulum lumenal Ca(2+)-binding protein grp78, Heat shock 70 kDa protein 5, Immunoglobulin heavy chain-binding protein, BiP) is encoded by the HSPA5 (also known as GRP78) gene (Gene ID: 3309) in human. GRP78 a member of the HSP family of molecular chaperones that is required for stress-induced autophagy and for endoplasmic reticulum (ER) integrity. GRP78 is involved in multiple processes, including translocation of the newly formed proteins across the ER membrane, folding and assembly of proteins, and targeting misfolded proteins for ER-associated degradation. It plays a critical role in regulating unfolded protein response (UPR). Following ER stress, GRP78 is released from ER transmembrane signal transducers, such as PKR-like ER kinase (PERK), which leads to the activation of UPR signaling pathways. Once GRP78 is dissociated, PERK is allowed to dimerize and is autophosphorylated and activated. Activated PERK phosphorylates elF2alpha that results in the reduction of protein synthesis. Specificity

This polyclonal antibody detects GRP-78 protein in multiple mammalian species. It targets an epitope within 17 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of human GRP-78.

Application

Anti-GRP-78, Cat. No. ABN1464, is a highly specific rabbit polyclonal antibody that targets 78 kDa glucose-regulated protein and has been tested in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected GRP-78 in human cerebellum, human pancreas, human small intestine, and human tonsil tissues.

Immunocytochemistry Analysis: A 1:50 dilution from a representative lot detected GRP-78 in NIH/3T3 cells.


Quality

Evaluated by Western Blotting in A431 cell lysates.

Western Blotting Analysis: 0.2 µg/mL of this antibody detected GRP-78 in 10 µg of A431 cell lysates.
