Specificity

Clone 4H18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects TGF-β receptor type 1 (TGF β-R1). It targets an epitope within 16 amino acids from the extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the extracellular domain of human TGF-β receptor type-1 (TGF β-R1).

Application

Quality Control Testing

Evaluated by Western Blotting with IgG-His-tagged recombinant protein fragment from the extracellular domain of Human TGFβ receptor 1.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IgG-His-tagged recombinant fragment corresponding to 100 amino acids from the extracellular domain of Human TGFβ receptor 1.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TGF β receptor 1 in HepG2 cells.

Affinity Binding Assay: A representative lot of this antibody bound TGF β receptor 1 peptide with a KD of 1.2 x 10-7 in an affinity binding assay.

Enzyme Immunoassay (ELISA) Analysis: Serial dilutions from a representative lot detected hIgG-His-tagged recombinant fragment corresponding to 100 amino acids from the extracellular domain of Human TGF β receptor 1.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

TGF-beta receptor type-1 (UniProt: P36897; also known as EC:2.7.11.30, TGFR-1, Activin A receptor type II-like protein kinase of 53kD, Activin receptor-like kinase 5, ALK-5, Serine/threonine-protein kinase receptor R4, SKR4, Transforming growth factor-beta receptor type I, TbetaR-I) is encoded by the TGFBR1 (also known as ALK5, SKR4) gene (Gene ID: 7046) in human. TGF-β receptor type-1 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-33), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 34-126), a transmembrane domain (aa 127-147), and a cytoplasmic domain (aa 148-503). It is widely expressed and is most abundant in placenta and least abundant in brain and heart. It is also expressed in a variety of cancer cell lines. TGF-β signaling involves its binding to the TGF-β receptor type II (TGF-β RII), which allows it to recruit TGF-β receptor type I (TGF-β RI) and assemble it as a heterod

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
