General description
Aprotinin from bovine lung is a globular polypeptide monomer with a molecular weight of 6.5 kDa. βα I residui di aprotinina dagli amminoacidi 13 - 18 sono essenziali per il legame alle serina proteasi.[1]
Application
L′Aprotinina è prevalentemente usata come inibitore della tripsina.
Aprotinina da polmone bovino è stata usata:
- come inibitore di proteasi in tamponi per radioimmunoprecipitazione (RIPA) per la omogeneizzazione di cellule cardiache microvascolari endoteliali (CMECs)(4) e cellule mammarie epiteliali
- in prove di angiogenesi di
- fibroblasti nella stabilizzazione proteomica della saliva surnatante.
Biochem/physiol Actions
L′Aprotinina è un inibitore competitivo di proteasi seriniche che forma complessi stabili con i siti attivi degli enzimi bloccandoli. Il legame è reversibile e la maggior parte dei complessi aprotinina-proteasi si dissocia a pH >10 o < 3. Strutturalmente l′Aprotinina è una proteina globulare monomera derivata da polmone bovino consistente in 58 aminoacidi, disposti in una catena polipetidica singola con tre ponti disolfuro.
La Aprotinina inibisce proteasi come tripsina, plasmina, chimotripsina e trombina. Blocca la sintesi di bradichinina da chininogeni. Utile per curare perdite di sangue in chirurgia.[1]
Preparation Note
Questo prodotto è una polvere liofilizzata con attività di 3-8 TIU/mg di polvere solida. La Aprotinina è molto solubile in acqua (>10 mg/mL) e in tamponi acquosi poco ionici. Le soluzioni diluite sono tendenzialmente meno stabili delle concentrate benché la stabilità della soluzione dipenda anche dal pH. La Aprotinina è relativamente stabile alla denaturazione - soltanto la termolisina si è trovata in grado di degradarla a 60-80 °C. La sterilizzazione della Aprotinina si può ottenere per filtrazione attraverso un filtro da 0,2 μm.
Other Notes
Una unità di inibitore di tripsina (TIU) diminuirà l′attività di due unità di tripsina del 50%, in cui una sola unità di tripsina idrolizzerà 1,0 μmoli di N-alfa-benzoil-DL-arginina p-nitroanilide per minuto a pH 7,8 e 25 °C. Un′altra unità usata comunemente è KIU, con 1 TIU = 1,300 KIU.
