General description

Monoclonal Anti-Thimet Oligopeptidase 1 (THOP1) (mouse IgG2b isotype) is derived from the hybridoma IVD6 produced by the fusion of mouse myeloma cells (SP2/O cells) and splenocytes from CF1mice immunized with a synthetic peptide. THOP1 protein is found in tissues with high levels of neuropeptides and hormones. THOP1 gene is mapped on the human chromosome at 19p13.3.

Specificity

Monoclonal Anti-Thimet Oligopeptidase recognizes human and monkey THOP1, ~85 kDa.

Immunogen

synthetic peptide corresponding to amino acids 47-63 of human THOP1.

Biochem/physiol Actions

Thimet Oligopeptidase 1 (THOP1) is a metalloendopeptidase found in the brains of Alzheimer patients. It can cleave a synthetic peptide encompassing the amino-terminus of amyloid β-peptide and generate amyloidogenic fragments from recombinant human amyloid precursor protein (APP). THOP1 is involved in degrading peptides that are released from the proteasome and therefore limits the extent of major histocompatibility complex (MHC) class I antigen presentation. It is involved in the processing of bioactive peptides. Overexpression of the gene THOP1 in cells reduced the presentation of antigenic peptides by class I MHC molecules. Prevention of THOP1 expression by small interfering RNA promoted class I antigenic peptide presentation.

Target description

THOP1 is a metalloprotease that is able to cleave a synthetic peptide encompassing the amino-terminus of amyloid b-peptideand generate amyloidogenic fragments from recombinant human Amyloid Precursor Protein.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in "frost-free" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
