General description

Triton X-100 is a common non-ionic surfactant and emulsifier which is often used in biochemical applications to solubilize proteins. It is considered a comparatively mild detergent, non-denaturing, and is reported in numerous references as a routinely added reagent. It is utilized for lysing cells to extract protein and cellular organelles. It can also permeabilize the living cell membrane for transfection.[1][2]

Application

Triton X-100 has been used:
  • Along with Span® 80 and Tween® 80 in mineral oil for the preparation of oil-phase of microemulsions for PCR technique[3][4][5]
  • As a component of NX buffer which is used to pool and break down the microemulsions after PCR cycling[3]
  • In the blocking solution during immunocytochemistry procedure[6][7]
  • During deglycosylation procedure[8]
  • For membrane solubilization[9]
  • For cell permeabilization[9][10]
  • In decellularization of porcine heart valve tissue[11]

Questions

1–8 of 8 Questions
  1. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  6. What is the Surface Tension value of Triton X-100? and, what is the hydrophilic/lipophilic balance (HLB) and critical micelle concentration (cmc) of Triton X-100?

    1 answer
    1. • Cloud Point @1% Aqueous (ASTM D 2024): 66 °C
      • CMC (25°C): 189 PPM
      • Foam Height - Initial (0.1 wt% actives): 128 mm
      • Foam Height-5 minutes (0.1 wt% actives): 107 mm
      • HLB: 13.4 Moles EO: 9.5 Pour Point: 1 °C

  7. What is the expiration date for the products?

    1 answer
    1. This product is not assigned an expiration date. It is assigned a recommended retest date of approximately 2.5 years after the quality release date. A recommended retest date is the period of time during which the product is expected to remain within established stability specifications, provided that it has been stored under defined conditions. After the Retest Date, product samples should be examined to ensure that the product is still in compliance with the established specifications. For more information, you may access the "Product Dating Information" document under "ADDITIONAL USEFUL DOCUMENTS ABOUT OUR PRODUCTS" at the bottom of the Quality Services page with this link: https://www.sigmaaldrich.com/US/en/life-science/quality-and-regulatory-management/quality-services.

  8. Hello, I bought the following product: Triton X-100 (T9284) - BioXtra and I was wondering what grade is it? Is it suitable for nucleic acid extraction? Thanks, Irene

    1 answer
    1. The BioXtra products are characterized by more stringent testing protocols that may include trace metal analysis and specialized suitability testing. This product is not tested specifically for nucleic acid extraction. However, there is no specific aspect of the product that would render it unsuited for this application. For future reference, the molecular biology grade (T8787) may be of interest. This product is tested for DNAse and RNAse activity. See the links below to review this product option:
