General description

PSIP1, also known as coactivator p75, as well as numerous other synonyms named after their original discoveries including, PC4 and SFRS1 interacting protein 1, lens epithelium-derived growth factor (LEDGF/p75), dense fine speckles 70kD protein (DFS 70), and transcriptional coactivator p75/p52, is a transcriptional coactivator involved in neuroepithelial stem cell differentiation and neurogenesis. It is particularly important in lens epithelial cell gene regulation and stress responses, and even though initially thought to be limited to the lens of the eye, PSIP1 is now known to be widely expressed in most tissues and has no direct role in the development of lens epithelium. It also gained research interest as the focal point for HIV integration and appears to be one of the few integrase interactor proteins whose knockdown limits HIV integration and propagation. Moreover, PSIP1 is an anti-apoptotic protein and appears to play a protective role during stress-induced apoptosis. There are three isoforms of the PSIP1 in humans, PSIP1, PSIP2 or p52, and a third isoform which is unnamed. The PSIP1 isoforms seem to vary in their precise functions somewhat with isoform 2 being the most commonly expressed and most abundant. Expression is localized to the nucleus. EMD-Millipore’s anti-PSIP1mouse monoclonal, clone 6.00E+04 has been tested on human, paraffin embedded immunohistochemical sections, immunocytochemical immunofluorescence studies using mouse NIH3T3 cells, western blot using human, mouse, and rat cells line lysates, and ELISA using purified antigen.

Application

Detect PSIP1 using this mouse monoclonal antibody, Anti-PSIP1 Antibody, clone 6E4 validated for use in western blotting, IHC & Immunofluorescence.
Immunofluorescence Analysis: A 1:200-1,000 dilution from a representative lot detected PSIP1 in NIH/3T3 cells.

Immunohistochemistry Analysis: A 1:1,000 dilution of this antibody detected PSIP1 in rat cerebellum, rat hippocampus, and human cerebellum tissues.

Optimal working dilutions must be determined by end user.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
