General description

SMAD6 is the gene name for a protein that is a homolog of the fly gene Mothers against decapentaplegic, and in humans it is called Mothers against decapentaplegic homolog 6 (MAD homolog 6). Other names include SMAD family member 6 and Mothers against DPP homolog 6 and another gene name of MADH6. By whatever name, the protein is a transcription factor that binds to regulatory elements within the nucleus and mediates transcription. The effect is to act as a mediator of TGF-beta and BMP mediated anti-inflammatory signaling activity. SMAD6 suppresses IL1R-TLR signaling, thus preventing NF-kappa B activation and activation of proinflammatory genes. SMAD6 interacts with numerous other intracellular proteins including NEDD4L, STAMBP, PRKK, RNF111, and AXIN1. SMAD6 is expressed in a wide variety of tissues with higher levels being seen in lung and cardiac tissues. Recently SMAD6 has been shown to promote neuronal differentiation by inhibiting the Wnt/beta catenin signaling pathway in addition to inhibiting the BMP signaling. Finally in cancer SMAD6 via its suppression of TGF-beta induced growth inhibition can potentiate the transformed phenotype. Moreover SMAD6 is often over expressed in many cancers, which suppresses G0-G1 arrest and apoptosis which might otherwise normally occur, thus leaving SMAD6 playing a critical role in supporting cancer cell growth and survival. EMD-Millipore’s Anti-SMAD6 mouse monoclonal antibody has been tested in western blot against human A431, HeLa, and Jurkat cell lysates.

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
