General description

Phospholipase D (PLD) catalyzes the hydrolysis of the phosphodiester bond of glycerophospholipids to generate phosphatidic acid and choline. It is ubiquitously expressed and is implicated in immune function and cancer inhibition. Abnormalities in PLD expression have been associated with human cancers such as breast, renal, gastric and colorectal cancer.

Application

Phospholipase D (PLD) Activity Assay Kit has been used to measure PLD activity in the liver and lysosomal fractions of the mice liver.
Suitable for the detection of phospholipase D activity in biological samples. This kit can be used to study the effects of various compounds on phospholipase D activity.

Principle

In this assay, PLD hydrolyzes phosphatidylcholine to choline, which is determined using choline oxidase resulting in a colorimetric (570 nm)/fluorometric (λex = 530/λem = 585 nm) product, proportional to the PLD activity in the sample. Unit definition: one unit of PLD catalyzes the formation of 1 mmole choline per minute under the assay conditions (pH 7.4).
