General description

Fibroblast growth factor 2 (FGF-2) belongs to the family of heparin-binding growth factors. It is localized in the cells of the mesoderm and neuroectodermal origin and several tumor cells. FGF-2 exists in four isoforms. It is made up of four cysteine residues, many basic residues, and two active sites. FGF-2 protein is a trigonal pyramidal structure formed by twelve anti-parallel β-sheets. The FGF-2 chromosome is located on the human chromosome at 4q28.1.

Specificity

This antibody is highly specific for the non-denatured form of FGF-2/basic FGF from bovine, human, rat and mouse sources. Does not cross-react with bovine acidic FGF.
This antibody recognizes the non-denatured form of FGF-2/basic FGF; does not cross-react with bovine acidic FGF.

Application

Anti-FGF-2/basic FGF (neutralizing) Antibody, clone bFM-1 has been used in:

  • FGF2 neutralization assay (6 or 12 µg/mL)
  • in vitro tube formation assay (1000 ng/mL)
  • immunolabelling (1:200)

Biochem/physiol Actions

Fibroblast growth factor 2 (FGF-2) plays a role in vertebrate embryonic and fetal development. It acts as an inducer of angiogenesis and is a well-known mitogen (in vitro) for a variety of cells including fibroblasts and vascular endothelial cells. FGF-2 exerts its biological activity through high-affinity tyrosine kinase receptors and low-affinity heparan sulfate proteoglycans present on the cell surface. It exhibits autocrine effects on various cell functions involved in angiogenesis such as proliferation, integrin expression, migration, and proteinase production. FGF-2 participates in the growth, migration, differentiation, and survival of a variety of cells. The binding of FGF-2 with heparin protects FGF-2 from denaturation by heat and acid and proteolytic cleavage.

Quality

Routinely evaluated by neutralization of greater than 50% of growth of FBHE cells simulated with FGF-2/bFGF.

Neutralization of bFGF Activity Assay:
2.5 µg/mL of this lot of antibody neutralized greater than 40% of growth of FBHE cells simulated with 5 ng/mL of FGF-2/bFGF.

Storage and Stability

Stable for 1 year at -20ºC from date of receipt. Aliquot to avoid repeated freezing and thawing. For maximum recovery of the product, centrifuge the original vial after thawing and prior to removing the cap.
