LW-3-254, a derivative of MM0299, is an orally bioavailable and blood-brain barrier penetrant, selective and potent inhibitor of lanosterol synthase (LSS) that exhibits anti-glioblastoma activity. LW-3-254 stimulates 24(S),25-epoxycholesterol (EPC) synthesis in GBM tumors, which induces toxicity in glioma stem-like cells. It potently inhibits Mut6 (Glioblastoma stem-like cells) cell growth by depleting cellular cholesterol.
