General description

Succinate, or succinic acid, can be found in all plants and animal tissues. It is an intermediate in the citric acid cycle and plays an important role in intracellular energy generation. Succinate is widely used as a flavoring agent in the food, beverage, and pharmaceutical industries due to its low toxicity.

Application

The Succinate Assay Kit may be used for:
  • Diabetes Research
  • Food and Beverage Testing
  • Mitochondrial Disorder Research

Features and Benefits

Efficient and Precise: Detect Succinate levels ranging from 10 to 400 μM (colorimetric) and 2 to 40 μM (fluorimetric) in a 30 minute room temperature reaction, providing both convenience and sensitivity.

Compatibility with High-Throughput Systems: Easily incorporate our kit into high-throughput handling systems, ensuring smooth and accurate processing, enhancing efficiency in your laboratory workflow.

Suitability

Suitable for succinate determination in food, beverage, agricultural products, and other biological samples.

Principle

The Succinate Assay Kit provides a simple, one step assay for measuring succinate. In this assay succinate is converted to pyruvate which reacts with specific reagents and dye to form a colored product. The color intensity at 570 nm or fluorescence at λex/em = 530/585 nm of the reaction product is directly proportional to succinate concentration in the sample. Linear detection range 10 μM to 400 μM for colorimetric assays and 2 to 40 μM for fluorimetric assays.

We have previously described an evolutionarily selected Tibetan prolyl hydroxylase-2 (PHD2D4E;C127S) variant that degrades the hypoxia-inducible factor (HIFα) more efficiently and protects these highlanders from hypoxia-triggered elevation in haemoglobin concentration. Phospho-Akt1 (pAkt1) undergoes prolyl hydroxylation at Pro125 and Pro313 by the prolyl hydroxylase-2 (PHD2) in a reaction decarboxylating α-ketoglutarate (αKG). We investigated whether the αKG supplementation could inhibit Akt-mediated activation of platelets and monocytes, in vitro as well as in
