General description

The FLAG epitope, is an eight amino acid protein with an enterokinase-cleavage site.[1][2]
The rabbit Anti-FLAG polyclonal affinity antibody ANTI-FLAG recognizes the FLAG epitope located on FLAG fusion proteins. This antibody reacts with N-terminal, N-terminal-Met, and C-terminal FLAG fusion proteins.

Application

The antibody recognizes the FLAG epitope located on FLAG-tagged fusion proteins at the N-terminus or C-terminus, applying dot blot, immunoblotting[3][4], immunoprecipitation[3] and immunocytochemistry assays.

    1. This product has been recommended for use in immunofluorescence and indirect immunofluorescence at 5-10 μg/mL using 293T cells transfected with a plasmid encoding FLAG-JNK. It has not been tested for use in IHC applications. The end-user will need to optimize the assay. Please see the link below to review the product datasheet:

      In general, antibodies work well on 5-15 uM sections, for 30 uM sections, blocking /permeabilization steps and antibody dilution, the incubation time will need to be optimized by end user.     1. To reduce non-specific background on tissue staining, a small amount of Triton X-100 (Product No. X100 or T8787), can be added to the wash buffers and antibody incubation buffers. Try a Triton X-100 concentration of 0.1% to start. Tween 20 (Product No. P5927) can also be used.

  9. How much Product F7425, ANTI-FLAG® from rabbit, should be used in western blotting and ELISA applications?

    1. For a Western blot from a PAGE run (preferably) under denaturing conditions, a 1:400 dilution of this antibody should be used.  A 1-10 μg/ml concentration should be used when using thie antibody in an ELISA application.
