General description

OptiPrep Density Gradient Medium, also known as iodixanol, is a low osmolarity, non-ionic iodinated density gradient medium. It is a sterile and endotoxin-tested solution that comprises 60% (w/v) iodixanol in water, designed for the in vitro isolation of biological particles. It may be used for fractionation and separation of cell and subcellular organelles.[1]

Application

OptiPrep has been used to isolate molecules, cells, and subcellular organelles, including DNA, cytokines, lysosomes, endoplasmic reticulum, peroxisomes, T lymphocytes, and natural killer cells.[1] It has also been used for the following applications:

  • to investigate the efficiency of single-cell encapsulation.[1]
  • the gradient-based isolation of senescent cancer cells.[2]
  • isolation of non-enveloped virion isolation from bronchoalveolar lavage.[3]
  • isolation of lipoproteins and macromolecules
  • purification of virions
  • purification of neuron cells

  • It is biologically inert and does not interfere with assays for marker enzymes.[1]
  • It is non-ionic, non-toxic, and has low osmotic pressure.
  • It forms spontaneous gradients that are stabler and easier to prepare than sucrose.[2]

  6. Why choose Product D1556, OptiPrep, instead of other density gradients such as Percoll or Histopaque?

Percoll and Histopaque must be removed prior to use through washing.  If Percoll and Histopaque are not removed, these density gradients can be toxic to mammalian cells.  In comparison, OptiPrep has no potential for bioaccumulation and can be considered nontoxic to cells. OptiPrep also has lower viscosity as compared to polysucrose based products such as Histopaque or Ficoll.

    Expensive OptiPrep

    It worked as OptiPrep from other companies, but it's over-priced, in my opinion.
