General description

Chemokine receptor CCR6 plays an important role in adaptive immunity, recruitment of CD4+ T cells and dendritic cells to sites of epithelial inflammation. It is expressed on leukocyte subsets of T cells, B cells, immature dendritic cells, neutrophils, monocytes and NKT cells. CCR6 binds the chemokine, CCL20 (macrophage inflammatory protein-3α) and induces proinflammatory signals such as TNF-α, IFN-γ and toll-like receptor agonists that in turn activate ERK1/2 and p38 MAPK pathways. CCR6 has been linked to autoimmune diseases of multiple sclerosis, rheumatoid arthritis, psoriasis and atherosclerosis. Monoclonal Anti-CCR-6 reacts with CCR-6 transfected cells and not with the parent cell line. The antibody shows no cross-reactivity with CCR-1, CCR-2, CCR-3, CCR-4, CCR-5, CCR-7, CXCR-1, CXCR-2, CXCR-3, CXCR-4 and STRL33 transfected cells.

Specificity

The antibody reacts with human CCR-6 transfectant cells and not the parent cell line. The antibody shows no cross-reactivity with CCR-1, CCR-2, CCR-3, CCR-4, CCR-5, CCR-7, CXCR-1, CXCR-2, CXCR-3, CXCR-4 or STRL33.

Immunogen

human CCR-6 transfected NSO mouse myeloma cells.

Application

Monoclonal Anti-CCR-6 antibody may be used for flow cytometry at a working concentration of 1μg/106 cells using human peripheral blood mononuclear cells. Antibody concentration of 25 μg/ml may be used for immunohistochemistry of frozen human tissue sections.

Physical form

Lyophilized from a 0.2 μm filtered solution in phosphate buffered saline with 5% trehalose.

Preparation Note

Purified using protein G.

Chemokines (chemotactic cytokines) drive and direct leukocyte traffic. New evidence suggests that the unusual CCR6/CCL20 chemokine receptor/ligand axis provides key homing signals for recently identified cells of the adaptive immune system, recruiting both pro-inflammatory and suppressive T cell subsets. Thus
