To form a complete medium, it should be combined with the basal medium: C-20211

Application

Keratinocyte Growth Medium 2 is a serum-free medium developed for the cultivation of epidermal keratinocytes without feeder cells. The medium is optimized for primary human cells but can also be used for horse and porcine keratinocytes, as well as mouse and rat keratinocytes after adapting the CaCl2 concentration. Quality

All lots of PromoCell Keratinocyte Growth Medium 2 are subjected to comprehensive quality control tests using primary human keratinocytes. Each lot is checked for growth promoting activity, adherence rate, and typical morphology of the tested keratinocytes. Approved inhouse lots of media are used as a reference. In addition, all lots of media have been tested for the absence of microbial contaminants (fungi, bacteria, mycoplasma).

Preparation Note

Final supplement concentrations after addition to the basal medium:Bovine Pituitary Extract: 0.004 ml / mlEpidermal Growth Factor (recombinant human): 0.125 ng / mlInsulin (recombinant human): 5 µg / mlHydrocortisone: 0.33 µg / mlEpinephrine: 0.39 µg / mlTransferrin (recombinant human): 10 µg / mlCaCl2: 0.06 mMNote: CaCl2 is provided as separate stock solution. This allows users to adjust the CaCl2 concentration individually. To ensure the maximal growth performance of normal human Keratinocytes, PromoCell recommends adding the entire 60 μlCaCl2 stock solution, which results in final 0.06 mM CaCl2 concentration. In case a higher CaCl2 concentration is required (e.g. differentiation experiments), a 0.5 M CaCl2-Solution can be ordered separately (C-34006).

Storage and Stability

Store the SupplementPack at -20°C in the dark immediately after arrival. If stored properly, it is stable until the expiry date stated on the label. After adding the supplements to the Basal Medium, the shelf life of the complete medium is 6 weeks at 4 to 8°C. Do not freeze the complete medium. For use, pre-warm only an aliquot of the complete medium and keep the remaining medium refrigerated at 4 to 8°C.
