General description

Zinc finger and SCAN domain containing protein 4, also called Zscan4, is an embryonic stem (ES) cell-specific transcription factor required to regulate pluripotency. Zscan4 binds and regulates telomere elongation in order to maintain genomic stability and acts as an activator of spontaneous telomere sister chromatid exchange (T-SCE) in undifferentiated ES cells.

Immunogen

KLH-conjugated linear peptide corresponding to mouse Zscan4.

Application

Anti-Zscan4 Antibody is a highly specific rabbit polyclonal antibody, that targets Zinc Finger Protein & has been tested in western blotting, Immunofluorescence & IP.
Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected Zscan4 in mouse iPS and R1 Embyonic Stem cells. Data courtesy of the Melanie Welham lab at The University of Bath, UK
Immunoprecipitation Analysis: A 1:4,000 dilution from a representative lot detected Zscan4 in cytoplasmic and nuclear extracts in IP experiments conducted by the Melanie Welham lab at The University of Bath, UK. (Data not shown)

Quality

Evaluated by Western Blotting in mouse Embryonic Stem cell lysates.

Western Blotting Analysis: 0.2 µg/mL of this antibody detected Zscan4 in 10 µg of Embryonic Stem cell lysates.

Target description

~65 kDa observed. The calculated molecular weight of this protein is 57 kDa An uncharacterized band may be observed at ~50 kDa in some cell lysates.ZSCAN4 bands may be observed at both ~72 kDa and ~40 kDa when preparing cell lysates with SDS buffer. Additionally, a ZSCAN4 doublet may be observed at ~72 kDa when using cytosolic or nuclear extracts.

Physical form

Cloned animals generated by somatic cell nuclear transfer (SCNT) have been reported for many years; however, SCNT is extremely inefficient, and zygotic genome activation (ZGA) is required for SCNT-mediated somatic cell reprogramming. To identify candidate factors that facilitate ZGA in

Unlike pluripotent cells, which generate only embryonic tissues, totipotent cells can generate a full organism, including extra-embryonic tissues. A rare population of cells resembling 2-cell-stage embryos arises in pluripotent embryonic stem (ES) cell cultures. These 2-cell-like cells display molecular features

Totipotent cells hold enormous potential for regenerative medicine. Thus, the development of cellular models recapitulating totipotent-like features is of paramount importance.
