General description

PI3 Kinase is a complex of N-terminal 6His-tagged, recombinant, full-length human p110α containing the mutation E545K and untagged, recombinant, full-length human p85α.

Biochem/physiol Actions

p110α (PIK3CA)/p85α (PIK3R1) isoform of phosphoinositide 3-kinase (PI3K), plays a critical role in the regulation of various cellular processes. These include cellular survival, proliferation, migration, invasion, and metastasis. Additionally, the signaling pathway involving the p110α isoform of PI3K is responsible for triggering physiological cardiac hypertrophy induced by exercise. PIK3 also plays a vital role in activating Protein kinase B or Akt (PKB/Akt), an essential component of insulin and growth factors signaling pathways. Furthermore, PIK3 participates in synthesizing PIP3 (phosphatidylinositol (3,4,5)-trisphosphate), a key signaling molecule within the PI3K/Akt pathway. These significant functions highlight the critical role of PIK3 in mediating important cellular signaling processes, including inositol phosphate metabolism. PIK3CA mutations can alter the tumor immunogenicity, which potentially affects its susceptibility to immunotherapies. The E545K substitution is a somatic mutation in p110α that has been associated with tumors of the colon and brain. Combined in vitro and in vivo studies have shown that this mutation confers higher lipid kinase activity than the wild type and is able to induce oncogenic transformation.

Storage and Stability

Store at -70°C from date of shipment. For maximum recovery of product, centrifuge original vial prior to removing the cap.

Rapidly thaw the vial under cold water and immediately place on ice. Aliquot unused material into pre-chilled microcentrifuge tubes and immediately snap-freeze the vials in liquid nitrogen prior to re-storage at -70°C.
