Specificity
Clone 1B6 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Carbonic anhydrase 9 (CA9). It targets an epitope within 24 amino acids from the cytoplasmic domain in the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal, cytoplasmic domain of human Carbonic anhydrase 9 (CA().
Application
Quality Control Testing
Evaluated by Western Blotting in Rat colon tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CA9 in Rat colon tissue lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CA9 in Human stomach tissue lysate.
Affinity Binding Assay: A representative lot of this antibody bound CA9 with a KD of 9.2 x 10-8 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CA9 in Human stomach tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CA9 in A431 cells.

Target description
Carbonic anhydrase 9 (UniProt: Q16790; also known as EC:4.2.1.1, Carbonate dehydratase IX, Carbonic anhydrase IX, CA-IX, CAIX, Membrane antigen MN, P54/58N, Renal cell carcinoma-associated antigen G250, RCC-associated antigen G250, pMW1) is encoded by the CA9 (also known as G250, MN) gene (Gene ID: 768) in human. Six evolutionary distinct classes of carbonic anhydrases (CA) have been described (α, β, γ, δ, ζ and η). However, only the α-class is expressed in mammals. Sixteen different α-CA isozymes have been described that vary in their catalytic activity and subcellular localization. Out of the twelve catalytically active forms five are cytosolic, four are membrane bound with their catalytic domain facing the extracellular space, two are mitochondrial, and one is secreted isoform. They catalyze the conversion of CO2 to the bicarbonate iOn™ and protons to mediate pH and CO2 homeostasis. CA9, a membrane bound isoform, is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-37), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 38-414), a transmembrane domain (aa 415-435), and a short cytoplasmic domain (aa 436-459). It′s α-carbonic anhydrase domain is localized in amino acids 139-390. In addition, it features a proteoglycan-like domain (PG) in its N-terminal region (aa 38-112) that is unique to CA9 and contributes to the assembly of focal adhesion contacts during cell migration and may function as a proton buffer to support its catalytic activity. CA9 expression in normal healthy tissues is restricted to epithelial cells in the stomach and gut but is strongly upregulated in many tumor tissues. Its expression is under the control of hypoxia-inducible factor 1 (HIF-1). CA9 expression is observed in hypoxic and mild hypoxic regions. Its expression is reported in a breast and colorectal cancer, glioblastoma, lung cancer, and cervical squamous cell carcinomas, and is usually linked to poor prognosis. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity™, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
