General description

Purified endo-β−(1→4)-xylanase from Thermomyces lanuginosus produced by submerged fermentation of a genetically modified Aspergillus oryzae microorganism
Studies show that the signal sequence found in the xylanase gene is critical for protein production in E. coli. Loss of this sequence resulted in a significant reduction in expression. [1]

Xylanase catalyses the degradation of xylan into xylooligosaccharides and D-xylose. It also breaks down hemicellulose. Xylanase is used for the production of biofuels, such as ethanol and xylitol. It improves the quality of bread and juices.[2] Xylanase is used in biobleaching process and decreases environmental pollution.[3]

Application

Xylanase has been used:
  • in the isolation of hemicellulose and production of oligosaccharides[4]
  • in viscosimetric assay[5]
  • in the comparison of commercial enzymes for the aqueous enzymatic extraction of corn oil from corn germ[6]
  • in the hydrolytic degradation of xylan[7]

  1. Will this enzyme in buffer be deactivated completely at 100°C boiling for 10 mins towards end of protocol or it needs acid treatment strictly ?

    1. We have not tested boiling this enzyme at the end of the protocol, we have only used acid.

  2. Is this enzyme powder soluble in water ? seems like particle dispersing in buffer at Ph 4.8 ?

    1. The active components of xylanase X2753 are readily soluble in water at concentrations that occur in normal usage. However, water solutions will be turbid because of the wheat flour used for standardization of the enzyme.

    1. Each Lot is guaranteed to have a specific activity greater than or equal to 2500 Units/g. There is no maximal value specified.

  4. What is the optimal pH, buffer, temperature and concentration of this product ?

    1. This is a product of Novozymes Corp. The manufacturer tests this material according to the following protocol and conditions:
      Temperature - 50°C
      Buffer - 0.1M Phosphate Buffer, pH 6
      Enzyme concentration - 0.007 - 0.057 units/mL
      Substrate - 0.45%w/v Azo-Wheat arabinxylan in 0.1 M Phosphate Buffer, pH 6
      Reaction - 0.100 mL enzyme solution added to 0.900 mL substrate solution
      Incubation time - 30 minutes in a water bath at 50°C
      Stop Reaction - add 5 mL of Stop Reagent: 6.66 mL 2 N HCl in 1000mL absolute Ethanol
      Incubation - 15 - 60 minutes at ambient temperature
      Centrifuge at 4000 rmp for 15 minutes
      Detection - 585 nm (manual) within 20 minutes of centrifugation
