General description

The new Fumarase Assay Kit, MAK521, is now available! Fumarase (Fumarate Hydratase; EC 4.2.1.2) catalyzes the reversible hydration of fumarate to malate in the tricarboxylic acid (Krebs) cycle. The activity of fumarase is important as it produces reducing equivalents that drive the synthesis of ATP and maintains energy balance in aerobic metabolism. A deficiency of fumarase activity results in progressive encephalopathy.

Fumarase Activity Colorimetric Assay Kit has been used to determine fumarase (FUM) activity.

Suitability

Suitable for the detection of Fumarase in a variety of biological samples such as cell and tissue culture supernatants and purified mitochondria.

Principle

The Fumarase Activity Colorimetric Assay Kit provides a simple and high-throughput ready procedure for measuring fumarase activity in a variety of tissues, cells, and isolated mitochondria. Fumarase activity is determined by measuring a colorimetric product with absorbance at 450 nm (A450) proportional to the enzymatic activity present. One unit of fumarase is the amount of enzyme that generates 1.0 mmole of NADH per minute at pH 9.5 at 37 °C.
