General description

Mindbomb homolog 2 (MIB2) which is also known as skeletrophin, is an ubiquitin ligase. The gene encoding it is localized on human chromosome 1p36.32.
Mouse MIB1 and MIB2 share 52% similarity in their amino acid sequences and contain a similar domain organization.These include two homologous to the E6-AP carboxyl terminus (HECT) domains. It also comprises RCC1-like domain (RLD) domain containing E3 ubiquitin protein ligase 2 (herc2)/mib domains.Other domains associated with MIB2 are one ZZ zinc finger, one mib repeat, ankyrin repeats, and two interesting new gene (RING) domains. MIB2 is highly expressed in adult tissues.

Specificity

Anti-MIB2 (C-terminal) specifically recognizes human and rat MIB2.

Immunogen

synthetic peptide corresponding to amino acides 950-966 of human MIB2 conjugated to KLH. The corresponding sequence differs by one amino acid in mouse.

Application

Anti-MIB2 (C-terminal) antibody produced in rabbit may be used in immunoblotting.

Biochem/physiol Actions

Disruption of the MIB2 gene results in exencephaly and appears to be less essential in mouse development.
Mindbomb homolog 2 (MIB2) takes part in the ubiquitination of Notch ligands like Jagged-2 (JAG2). It binds to actin and CYLD (cylindromatosis (turban tumor syndrome)), which is an ubiquitin hydrolase. It has also been shown that MIB2 negatively regulates the transcription of Met oncogene.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
