Specificity

The product is found to be reactive with Anti-Avidin, Catalog Number A5170, and non-reactive with Anti-Streptavidin, Catalog Number S6390, by immunoelectrophoresis.

Application

ExtrAvidin®-Peroxidase is suitable for the following applications:
  • Dot blot at a minimum dilution of 1:4,000
  • Immunohistochemistry (using formalin-fixed, paraffin-embedded sections at a dilution of 1:100)[1][2][3][4][5][6]
  • Direct ELISA at a minimum dilution of 1:2,000[7][8]
It has also been used for:
  • Immunoprecipitation and Immunobloting[9]
  • Ra2I solid-phase binding assays[10]
  • Southwestern blotting[11]
  • Western blotting[11][12][13]
  • Immunocytochemistry[14]

Biochem/physiol Actions

Avidin is a tetrameric or dimeric biotin-binding protein produced in the oviducts of birds, reptiles and amphibians and deposited in the whites of their eggs.[15] It consists of four high affinity binding sites for biotin. ExtrAvidin® is prepared from egg white avidin. It is a modified form of affinity purified avidin with high specific activity of avidin and low background staining of streptavidin. It is a biotin binding protein produced by the bacteria Streptomyces avidinii.[16] ExtrAvidin-Peroxidase may be used in conjunction with biotin in different techniques.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 0.05% MIT

Storage and Stability

Store at 2-8 °C. Do Not Freeze. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
