Specificity

lone 1N18 is a is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Partitioning defective 3 homolog (PARD-3). It targets and epitope within the N-terminal half.

Immunogen

His-tagged recombinant fragment corresponding to 288 amino acids from the N-terminal half of human Partitioning defective 3 homolog (PARD-3).

Application

Quality Control Testing

Evaluated by Western Blotting in U2OS cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected PARD-3 in U2OS cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected PARD-3 in HeLa cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound PARD-3 with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PARD-3 in U2OS cells.

Anti-PARD-3, clone 1N18 ZooMAb®, Cat. No. ZRB1331, is a recombinant Rabbit monoclonal antibody that specifically targets PARD-3 and is tested for use in Affinity Binding Assay, Immunocytochemistry, and Western Blotting.

Target description

Partitioning defective 3 homolog (UniProt: Q8TEW0; also known as PAR-3, PARD-3, Atypical PKC isotype-specific-interacting protein, ASIP, CTCL tumor antigen se2-5, PAR3-alpha) is encoded by the PARD3 (also known as PAR3, PAR3A) gene (Gene ID: 56288) in human. PARD-3 is an adapter protein that is involved in asymmetrical cell division and cell polarization processes and plays a central role in the formation of epithelial tight junctions. It is localized along the cell-cell contact region. PARD-3 is shown to inhibit atherosclerosis onset by blocking endothelial inflammation. It also colocalizes with the cortical actin that overlays the meiotic spindle during metaphase I and metaphase II. In its N-terminal half, it contains three PDZ domains and a conserved oligomerization domain that is shown to be essential for proper subapical membrane localization. Its second PDZ domain (aa 461-546) mediates interaction with membranes containing phosphoinositol lipids. Together with PARD6, atypical protein kinase C (aPKC), and the Rho GTPase CDC42, it forms a complex that regulates cytoskeleton re-organization, post-Golgi vesicle trafficking, microtubule network polarization, and asymmetric cell division. PARD-3 can undergo phosphorylation at serine 827 by PRKCZ and PRKCI at the most apical tip of epithelial cell-cell contacts during the initial phase of tight junction formation and promote dissociation of the complex with PARD6. Mutations in PARD3 gene cause neural tube defects leading to congenital malformations of the central nervous system. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
