General description

Stimulated by retinoic acid gene 8 protein (UniProt P70278) is encoded by the Stra8 gene (Gene ID 20899) in murine species. Stra8 is an early retinoic acid (RA) responsive gene that encodes a cytoplasmic protein expressed exclusively in gonads, where it plays a pivotal role in initiating meiosis of both female and male germ cells. In female mice, Stra8 is expressed in embryonic gonocytes during a restricted time window and is indispensable for their entry into the meiotic S phase. In males, Stra8 expression is first detected in primitive spermatogonia at P5, then becomes prominent in premeiotic (preleptotene) spermatocytes from P10 onwards. Spermatogenesis, the transformation of stem cells into millions of haploid spermatozoa, is a highly regulated process that allows a constant spermatozoa production. Undifferentiated mitotic spermatogonia first develop into committed spermatogonia to produce spermatocytes, which then undergo meiosis to form haploid spermatids. Stra8 is not only required for meiosis, but also promotes spermatogonial differentiation. In the absence of Stra8, spermatogonial differentiation is impaired, giving rise to massive accumulations of undifferentiated and early differentiating spermatogonia (Type A spermatogonia) in aged testes. Stra8-deficient germ cells in postnatal male and fetal female mice arrest just before meiosis, without entering meiotic prophase.

Specificity

This polyclonal antibody detected Stra8 immunoreactivity in testes from wild-type, but not Stra8-knockout mice (Mark, M., et al. (2008). J. Cell Sci. 121(Pt 19):3233-3242).

Application

Anti-Stra8 Antibody, Cat. No. ABN1656, is a highly specific rabbit polyclonal antibody that targets STRA8 and has been tested in Electron Microscopy, Immunocytochemistry, Immunofluorescence, Immunohistochemistry, and Western Blotting.
Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected Stra8 immunoreactivity in human testis tissue section.

Western Blotting Analysis: 2 µg/mL from a representative lot detected an upregulation of ~35 kDa Stra8 isoform in 10 µg of lysate from retinoid acid-treated P19 murine embryonic carcinoma cells (Courtesy from the laboratory of Dr. Mustapha Oulad-Abdelghani, IGBMC, France).

Electron Microscopy Analysis: A representative lot detected Stra8 immunoreactivity in the cytoplasm of germ cells in direct contact with the basal lamina and displaying features of spermatogonia. No Stra8 immunoreactivity was found in Sertoli cells in mouse testis cryosections (Oulad-Abdelghani, M., et al. (1996). J. Cell Biol. 135(2):469-477).

Immunocytochemistry Analysis: A representative lot detected exogenously expressed murine Stra8 in 2% paraformaldehyde-fixed, 0.1% Triton X-100-permeabilized COS-7 transfectants (Oulad-Abdelghani, M., et al. (1996). J. Cell Biol. 135(2):469-477).

Immunofluorescence Analysis: A representative lot detected stage-dependent Stra8 immunoreactivity among A1 A4 differentiated spermatogonia, preleptotene spermatocytes, and in early meiotic spermatocytes by fluorescent immunohistochemistry staining of sections from paraformaldehyde-perfused mouse testis (Mark, M., et al. (2015). Biochim. Biophys. Acta. 1849(2):84-93).

Immunofluorescence Analysis: A representative lot detected Stra8 immunoreactivity in 4% paraformaldehyde-fixed testes sections from wild-type, but not Stra8-knockout mice (Mark, M., et al. (2008). J. Cell Sci. 121(Pt 19):3233-3242).

Immunohistochemistry Analysis: A representative lot detected Stra8 immunoreactivity restricted to cells within the peripheral layer of the tubule of mouse testis cryosections. No Stra8 immunoreactivity was found in Sertoli cells (Oulad-Abdelghani, M., et al. (1996). J. Cell Biol. 135(2):469-477).

Western Blotting Analysis: A representative lot detected an induction of Stra8 expression in multiple phosphorylated forms in P19 murine embryonic carcinoma cells upon stimulation by either all-trans (T-RA) or 9-cis (9C-RA) retinoic acid (Oulad-Abdelghani, M., et al. (1996). J. Cell Biol. 135(2):469-477).

Western Blotting Analysis: 1 µg/mL of this antibody detected Stra8 expression in 10 µg lysate from all-trans retinoic acid-treated P19 murine embryonic carcinoma cells.

~53/35 kDa observed. 36.91/44.56 kDa (human/mouse; UniProt Q7Z7C7/P70278) calculated. Note: At least five spliced isoforms ranging from 35 to 55 kDa have been identified. Uncharacterized bands may be observed in some lysate(s).
