Specificity

Cyclooxygenase-2. Two isoforms of Prostaglandin H synthase are well characterized, namely COX1 and COX2. Both forms of COX proteins are membrane associated heme proteins containing cyclooxygenase and peroxidase activities. These enzymes are targets of NSAID (Non steroidal anti-inflammatory drugs) such as aspirin. COX2 is a 72 kDa protein having 60% homology to COX1. It is induced by cytokines and mitogens and likely plays a role in inflammatory diseases such as rheumatoid arthritis.

Immunogen

17 amino acid synthetic peptide corresponding to a sequence near the C-terminus of Human/Mouse/Rat COX-2 coupled to KLH.

This Anti-Cyclooxygenase-2 Antibody is validated for use in ELISA, WB for the detection of Cyclooxygenase-2.
Western blot: 1-10 μg/mL using Chemiluminescence technique; COX-2 runs as a 68-72KDa protein in most cells, sometimes a 90kDa post-translationally modified form is seen as well. CaCo-2 line extracts (induced with serum or growth factors) are good positive controls.

ELISA: 1:10,000 - 1:100,000 using 100 ng of immunogen peptide per well.

Optimal working dilutions must be determined by end user.

Physical form

Affinity purified immunoglobulin. Liquid in PBS with 0.1% BSA.

Storage and Stability

Maintain frozen at -20°C in undiluted aliquots for up to 6 months after date of receipt. Avoid repeated freeze/thaw cycles.

During shipment, small volumes of product will occasionally become entrapped in the seal of the product vial. For products with volumes of 200 μL or less, we recommend gently tapping the vial on a hard surface or briefly centrifuging the vial in a tabletop centrifuge to dislodge any liquid in the container′s cap.
