General description

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

This antibody is expected to demonstrate broad species reactivity due to sequence conservation.
This antibody recognizes Histone H3 phosphorylated at Ser28.

Use Anti-phospho-Histone H3 (Ser28) Antibody (Rabbit Polyclonal Antibody) validated in WB, ICC, DB, PIA, ChIP to detect phospho-Histone H3 (Ser28) also known as H3F3, H3F3B, H3S28P.
Western Blot Analysis: A 1:2,000 dilution from a representative lot detected phospho-Histone H3 (Ser28) in 10 µg of untreated and colcemid treated HeLa acid extract.

Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected phospho-Histone H3 (Ser28) in HeLa cells.

Chromatin Immunoprecipitation Analysis: A representative lot immunoprecipitated phospho-Histone H3 (Ser28) in colcecmid treated HeLa cell lysate.

Dot Blot Specificity Analysis: A representative lot of this antibody detected peptides corresponding to phospho-Histone H3 (Ser28), but not peptides corresponding to other unmodified and modifed Histones.

Peptide Inhibition Analysis: A 1:500 dilution form a representative was blocked by phospho-Histone H3 (Ser28) peptides, but not non-specific peptides.

Quality

Evaluated by Western Blot in HeLa acid extract.

Western Blot Analysis: A 1:2,000 dilution of this antibody detected phospho-Histone H3 (Ser28) in 10 µg of HeLa acid extract.
