How stable is hydrocortisone? I use it and supplement a medium but I administer it every 48 hours, can I have my stock for 7 days and it does not lose its effect or do I have to defrost it every time I use it so that it does not lose its effect?
1 answer-
The stability of this product in solution has not been determined. To assure optimum performance, it is recommended that solutions be stored in frozen aliquots at -20°C. However, the water soluble version of Hydrocortisone, available as product H0396, is estimated to be stable for 1 - 2 weeks when refrigerated at 2 - 8°C. Note that a publication, J. AM. PHARM. ASSOC., 49, 253 (1960), determined that solutions prepared in ethanol at 40mg/mL and stored at 25°C decline by 50% in potency after 160 days.

Can it be dissolved completely in absolute ethanol, and stored per instructions (instead of 1:1 chloroform and ethanol)?
1 answer-
A stock solution can be prepared by reconstituting the product at 1 mg/ml in 100% EtOH. Gently swirl to dissolve. Then dilute to 50 ug/ml with sterile culture media, aliquot, and store at -20 °C. As a frozen aliquot, this should be stable for at least 3-4 months. Avoid the freeze/thaw cycle.
