General description

IL12 (interleukin 12), a member of IL-12 family, is an immunoregulatory cytokine. It is a heterodimer composed of p40 and p35 subunit linked covalently by disulfide bonds. It has a molecular weight of 70kDa. p40 subunit is copiously produced by phagocytic cells, whereas p35 is produced at low level ubiquitously and constitutively. This cytokine was first recognized in 1989 as a natural killer (NK) cell-stimulatory factor. It is predominantly expressed on antigen-presenting cells (APCs) including dendritic cells (DCs), monocytes, macrophages, and Toll-like receptor induced-B cells.
Recombinant murine IL-12 is a 75.0 kDa heterodimeric glycoprotein consisting of disulfide-linked 35 kDa (p35) and 40 kDa (p40) subunits (506 total amino acid residues).

Biochem/physiol Actions

IL-12 is a potent regulator of cell mediated immune responses and it induces IFN-g production by NK and T cells. Recombinant murine IL-12 is a 75.0 kDa heterodimeric glycoprotein consisting of disulfide-linked 35 kDa (p35) and 40 kDa (p40) subunits (506 total amino acid residues).
IL12 (interleukin 12) is crucial for the maturation of naive T cells into IFNγ (interferon)–producing Th1 (T helper) cells. IL12 mutant mice show higher susceptibility to bacterial infections due to significant reduction in the production of IFNγ. This protein functions as an early pro-inflammatory cytokine during infections. IL12 has potent anti-tumor effects in preclinical studies, which however, have not been reproduced in clinical studies. Studies in mice show that IL12 plays a crucial role in immune response against methicillin-resistant Staphylococcus aureus (MRSA)-mediated lung infection. Mice treated exogenously with IL12 prior to being subjected to MRSA infection show enhanced bacterial clearance and non-fatality.

Physical form

Lyophilized from 0.5x PBS, pH 7.2.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in 1x PBS, pH 7.2-7.4 to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
