Gene Information

Specificity

Clone WEN42 is a mouse monoclonal antibody that detects C-type lectin domain family 4 member D (CD368/CLEC4D). It targets an epitope within the extracellular domain.

Immunogen

BWZ.36 cells expressing chimeric CD368/CLEC4D

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant Rat CD368/CLEC4D protein.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected recombinant fragment of Rat CD368/CLEC4D expressed with Fc region of human IgG1.

Target description

C-type lectin domain family 4 member D (UniProt: Q69FH1; also known as C-type lectin superfamily member 8, CD368) is encoded by the Clec4d (also known as Clecsf8, Mcl) gene (Gene ID: 362432) in rat. CD368 is a membrane surface receptor that is expressed on blood monocytes and neutrophils, as well as on several tissue macrophage populations, including alveolar and peritoneal cavity macrophages. It is a single-pass type II protein that contains a cytoplasmic domain (aa 1-24), a transmembrane domain (aa 25-47), and an extracellular domain (aa 48-218). Its extracellular C-terminal region contains the C-type lectin-like domain (aa 89-206) that contains an evolutionarily conserved folded domain, and a carbohydrate recognition domain containing the Ca2+ binding sites. CD368 acts as a pattern recognition receptor (PRR) of the innate immune system that recognizes damage-associated molecular patterns (DAMPs) of pathogen-associated molecular patterns (PAMPs) of bacteria and fungi. It can form disulfide-linked heterodimer with C-type lectin domain family 4 member E (CLEC4E) and with CLEC6A that forms a PPR against fungal infections. (Ref.: Lobato-Pascual, A., et al. (2013).
