Each well was seeded with 10,000 cells in 10% FBS DMEM for 18 hours. Subsequently, the media was replaced with 1% FBS DMEM containing up to 1 M 2-Deoxy-D-Glucose. After 24 hours, the cell media was removed for the assay. Typically, each well is seeded with 100 µL of 10,000 cells.
