Application

Human AB serum is used in tissue engineering, transplantation and cell therapy applications for the expansion of mesenchymal stem cells (MSC) from adipose tissue or mesenchymal stromal cells from human bone marrow; for standardized limbal epithelial stem cell graft generation and transplantation; for ex vivo expansion of NK cells from peripheral blood in Hematopoeitic Stem Cell Expansion Medium; and for upgrading pretransplant human islet culture technology.

Packaging

Packaging may vary for different batches due to short supply of the bottles used to pack this product. The presence or lack of shrink wrap around the cap of the bottle doesn’t impact product integrity/sterility of the product.

Warning

Each donor is tested for and found non-reactive for Hepatitis B & C and non-reactive for Human Immunodeficiency Virus (HIV) antibody by ELISA. Nevertheless, products of human origin should be considered potentially infectious and handled accordingly.

It is not unusual for our human serum products to be cloudy. This product is from pooled serum and cloudiness indicates that lipids are present in the serum. No clear, lipid-free human serum products are available.

  1. What is the best way to thaw this product? Once thawed, how long can we use it if kept in at 4 oC?

    1. The recommended storage temperature is -20°C. The following steps confirm the best way to thaw serum:

      1. Remove the serum bottles from the freezer and allow them to acclimate to room temperature for approximately 10 minutes.
      2. Place each container in a 30 to 37 °C water bath or incubator. Excessive temperatures will degrade heat-labile nutrients. If using a
      water bath, prevent the bottle caps from being completely submerged.
      3. Gently swirl or shake the bottles every 10 - 15 minutes until the serum is completely thawed.
      Please note: Multiple thaw/freeze cycles should be avoided as they will hasten serum nutrient degradation and can induce insoluble precipitate formation.

  3. If the serum is being used to complete media for a cell thaw but then the cells are put through a DNA extraction rather than put into culture - would thawing a bottle of serum, aliquoting, and then re-freezing the serum be okay?

    1. Thawed serum should be mixed thoroughly before being added to cell culture medium. Repeated freeze-thaw cycles may significantly impact serum quality. It is recommended to aliquot the thawed serum for single use and store aliquots at -20 °C. If serum is stored between 2-8 °C, it should be used within 2-4 weeks.

  5. At which biosafety level should Human Serum H6914 be handled?

    1. Blood products should be treated and handled as potentially infectious. It is recommended to use human origin products from normal (non-diseased) donors at Biosafety Level 2 (BSL-2). Although donors may be tested for adventitious infections, as listed on the Certificate of Analysis for the product, no testing can cover all types of infection. Therefore, these products should be treated as potentially infectious and handled accordingly. Laboratory directors and principal investigators are responsible for performing a risk assessment and determining the biosafety level in accordance with applicable national and local regulations.

    1. The shelf life is not extended for those product which are assigned a firm expiration date. No further testing is performed on these lots.

  8. What is the difference between human serum from AB plasma and  human serum from AB clotted whole blood?

    1. Product H6914 is prepared from whole blood that is allowed to clot, centrifuged and the serum removed. There is no additional processing required.Product H4522 is prepared from anticoagulated (sodium citrate) whole blood. This is centrifuged to pellet the cells (RBC, WBC, some platelets) and the plasma is removed. This plasma is then treated with calcium to activate the clotting cascade. The preparation is centrifuged again and the supernatant is removed. This product can also be called plasma derived serum.Product H6914 will contain more growth factors since it is allowed to spontaneously clot and there will be the release of growth factors from many of the cells (WBC, platelets) present.  The cells are removed before the plasma is clotted for Product H4522, so this plasma will have fewer growth factors present (more similar to what is circulating).
