General description

El suplemento de medios selenito de insulina-transferrina-sodio respalda el crecimiento y la maduración in vitro de los folículos a partir de los ovocitos y se utiliza sistemáticamente en los métodos de fertilización in vitro .[1]
Las formulaciones exentas de suero y con cantidad reducida de suero suelen requerir formas purificadas de los factores normalmente suministrados por el suero que son adecuadas para estudios de crecimiento y diferenciación in vitro. Estos factores importantes son: insulina, transferrina, selenio, piruvato y etanolamina.

La INSULINA es una hormona polipeptídica que promueve la captación de glucosa y aminoácidos y quizá deba a esta propiedad el efecto mitógeno observado.

La TRANSFERRINA es una proteína transportadora de hierro. El hierro es un oligoelemento esencial, pero puede ser tóxico en forma libre. Para nutrir las células en cultivo, se suministra unido a la transferrina en suero.

El SELENIO es un oligoelemento esencial que normalmente proporciona el suero.

Los estudios nutricionales indican que la mayoría de las células de mamíferos utilizan los componentes del suplemento descritos. Mejoran la proliferación celular y disminuyen la necesidad de suero de muchos tipos de células. Cuando el suplemento proporcionado se utiliza con un 4 a 2 por ciento de suero, se comunica una proliferación similar a la obtenida con medio suplementado con un 10 por ciento de suero.

Application

El suplemento de medios compuesto por selenito de insulina-transferrina-sodio se ha utilizado como:
  • suplemento del medio 199 (M199) para el cultivo de cardiomiocitos[2]
  • componente del medio 1640 Roswell Park Memorial Institute (RPMI) para el cultivo de células madre pluripotentes (IPSC)[3]
  • suplemento del medio Eagle modificado de Dulbecco′(DMEM) en células de carcinoma de ovario ID8[4]

Other Notes

Cada vial contiene: ≥21 mg de insulina humana recombinante; ≥19 mg de transferrina humana (sustancialmente exenta de hierro) y 25 μg de selenito de sodio.
La ITS es una mezcla de insulina humana recombinante, transferrina humana y selenito de sodio. Se trata de un suplemento celular general diseñado para ser utilizado en medios no complejos (p. ej MEM, RPMI-1640) y medios complejos (p. ej, F-12 de Ham, DME/F-12, MEM) con piruvato de sodio.

  2. When we add this ITS at 100X to our Ham's F12 medium it drastically adjusts the color, and the pH drops to ~6.5 from 7.2. The ITS is made up exactly as directed with 5ml acidified water and 45ml sterile water and this still occurs, why? Medium is buffered

    1. The 100X stock solution of this product is prepared by reconstituting this product in 5 mL water containing 0.1 mL of Acetic acid. In general, culture media are buffered using Sodium Bicarbonate or other suitable buffers. While prepared in acidified water, the volume of the ITS stock solution added is typically insufficient to effect the pH of a buffered medium. The molarity of the acetic acid is not specified, which may be contributing to the drop in pH of the final medium. Reducing the molarity of the Acetic acid may resolve the issue, 1 M is commonly used. See the link below, at the bottom of page 1, for recommended preparation instructions.   3. The *I1884 needs to be prepared with acidified water, which acidifies the pH of the medium when it is added to the culture medium, is it necessary to adjust the pH of the I1884 to pH 7-7.4, after preparing it and before adding it to the medium?

    1. Adjusting the pH is not necessary or recommended. This is prepared at a 100X concentration. The volume added to buffered culture medium for the 1X amount is not sufficient to cause a shift in the pH.

  5. What is the stability of I1884, Insulin-transferrin-sodium selenite media supplement (ITS Supplement), in solution?

    1. The stock solution can be frozen in working aliquots. Avoid repeated freeze/thaw cycles. Frozen working aliquots will be stable for at least one year. The solution is stable at 2-8°C for 30 days.

  6. How should I reconstitute I1884, Insulin-transferrin-sodium selenite media supplement (ITS Supplement)?

    1. First, make acidified water by adding acetic acid to reduce the pH to approximately 2. To prepare a stock solution, dissolve the contents of the vial in 5 mL sterile, acidified water and swirl to dissolve. Then add 45 mL of sterile water. This prepared solution is at 100X.  It is diluted 100 fold (1 ml to 100 ml) to give a 1X final concentration.

    1. The starting material has an activity of greater than or equal to 25.2 USP units/mg insulin.  The concentration of insulin at 100X is 5 mg/L.
