General description

Anti-Palladin (C-terminal) is produced in rabbit using as the immunogen a synthetic peptide corresponding to a sequence at the C-terminal of human palladin, conjugated to KLH. Palladin, an actin associated Ig-domain protein is mapped to human chromosome 4q32. This phosphoprotein is widely expressed in vertebrate cells and tissues. Palladin is expressed as three major isoforms of 90, 140 and 200 kDa.

  • immunoblotting
  • immunoprecipitation
  • immunofluorescence

Rabbit anti-Palladin (C-terminal) antibody can be used for immunoprecipitation (10-20μL), indirect immunofluorescence (1:250-1:500) and western blot (1:250 -1:500) using HeLa cells.

Biochem/physiol Actions

Palladin is a component of actin-containing microfilaments that control cell shape, adhesion, and contraction. Its expression is required for normal mammalian embryogenesis since inactivation of paladin leads to embryonic lethality. Palladin interacts with the actin-associated proteins -actinin, VASP, ezrin, and profilin, suggesting that it is required for normal actin cytoskeleton organization. It serves as a transcriptional regulator, which controls the expression of smooth muscle cell genes.
Palladin is a microfilament-associated phosphoprotein that is known to colocalize with alpha-actinin at cell-cell junctions, stress fibres, embryonic Z-lines, and focal adhesions. Palladin abnormalities have been linked to familial pancreatic cancer. Rabbit anti-Palladin (C-terminal) antibody binds to human palladin. Detection of the palladin band by immunoblotting is specifically inhibited by the immunizing peptide. A nonspecific band of approx. 55kDa may be detected in some cell extract preparations.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
