General description

Stomatin (UniProt: P27105; also known as Erythrocyte band 7 integral membrane protein, Erythrocyte membrane protein band 7.2, Protein 7.2b) is encoded by the STOM (also known as BND7, EPB72) gene (Gene ID: 2040) in human. Stomatin is a major integral protein of erythrocyte membranes that is palmitoylated and forms homo-oligomers and associates with lipid rafts. It contains a cytoplasmic domain (aa 1-25); an intramembrane domain (aa 26-54); and a cytoplasmic domain (aa 55-288). Its amino acid residues 265-273 are essential for homooligomerization. Stomatin was named after the rare human haemolytic anaemia hereditary stomatocytosis. It is ubiquitously expressed in all tissues and higher expression is observed in hematopoietic cells. Lower expression is observed in the brain. It is known to associate with the plasma membrane and cytoplasmic vesicles of fibroblasts, epithelial and endothelial cells, and specialized endosomes/granules in hematopoietic cells. At its N-terminal region it contains a 24-residue, basic domain that is phosphorylated at serine 10 by a cAMP-dependent protein kinase. This is followed by a 29-residue hydrophobic domain that is associated with the cytoplasmic face of the membrane and is palmitoylated at Cys-30. This hydrophobic intramembrane domain (residues 26 54) contains the conserved residue Pro-47, which is essential for its rare monotopic membrane protein structure. Adjacent to the intramembrane domain is a short sequence, KIIKEYERAII (aa 55-65) that displays similarity to the cholesterol-binding CRAC (cholesterol recognition/interaction amino acid consensus) motif. In its structure palmitoylated Cys-87 is essential for correct membrane attachment. Stomatin is known to interact with various ion channels and modulates their activity. It is also reported to inhibit the activity of Akt, suppress cancer cell proliferation, enhance apoptosis in vitro and inhibit xenograft tumor growth in vivo.Two isoforms of stomatin have been described that are produced by alternative splicing. Specificity

Clone GARP-50 is a mouse monoclonal antibody that detects Stomatin (STOM). It targets an epitope within the N-terminal region.

Immunogen

Purified native Stomatin from human erythrocytes.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysates.

Western Blotting Analysis: A 1:500 dilution of this antibody detected STOM in HeLa cell lysates.
