General description

RNA polymerase II subunit B1 (RPB1) is the largest subunit of the RNA polymerase II complex. As a holoenzyme RNA polymerase II catalyzes transcription of eukaryotic DNA into RNA using the four ribonucleoside triphosphates as substrates. The RBP1 subunit, in combination with other polymerase subunits, forms a large central cleft that maintains contact between the active site of the enzyme, the DNA template, and the nascent RNA transcript. This subunit also contains a carboxy terminal domain (CTD) consisting of tandem heptapeptide repeats. In actively transcribing RNA polymerase ‘Ser-2’ and ‘Ser-5’ of the heptapeptide repeat are phosphorylated. Phosphorylation activates the RNA polymerase II beta subunit, allowing it to serve as an assembly platform for additional subunits that modulate initiation, elongation, termination and mRNA processing.

Specificity

This antibody recognizes RNA polymerase II subunit B1.

Immunogen

Epitope: Unknown
Ovalbumin-conjugated linear peptide corresponding to human RNA polymerase II subunit B1.

RNA polymerase II is distinguished by its large carboxyl-terminal repeat domain (CTD), composed of repeats of the consensus heptapeptide Tyr1-Ser2-Pro3-Thr4-Ser5-Pro6-Ser7. Differential phosphorylation of serine-2 and serine-5 at the 5' and 3' regions of genes appears to coordinate the localization of

BRD4, a Bromodomain and Extraterminal (BET) protein family member, is a promising anti-cancer drug target. However, resistance to BET inhibitors targeting BRD4 is common in solid tumors. Here, we show that cancer-associated fibroblast (CAF)-activated stromal signaling, interleukin-6/8-JAK2, induces BRD4 phosphorylation
