General description

Keratin 18 (KRT 18) or cytokeratin 18 (CK18) encodes the type I intermediate filament chain keratin 18. The antibody DC-10 reacts with Cytokeratin 18 (45 kDa). Cytokeratins are a member of intermediate filaments subfamily represented in epithelial tissues. In human chromosome, the gene KRT18 is localized on 12q13.13.

Application

The reagent is designed for Flow Cytometry. Suggested working dilution is 1:500. Indicated dilution is recommended starting point for use of this product. Working concentrations should be determined by the investigator.

Biochem/physiol Actions

Keratin 18 (KRT18/CK18) pairs with CK8 to form the intermediate filaments and maintain the integrity in hepatocytes. CK18 associates with histone H3 in the progression of hepatocellular carcinoma (HCC). Disorganization or altered levels of CK18 is related to plectin deficiency in HCC. K18 may be a marker for liver diseases. K18 is present in Mallory bodies and is elevated in alcoholic cirrhosis and alcoholic hepatitis. K18 expression is modulated by interleukin 6 (IL-6). K18 interacts with desmosomal protein/ E-cadherin and promotes cell adhesion. CK18 is cleaved by caspases during apoptosis and released into blood as caspase cleaved cytokeratin-18 (CCCK18).
