Gene Information

General description

Zinc finger protein 746, also known as PARIS, parkin interacting substrate (ZNF746 ) is a 644 amino acid protein. It has a Kruppel-associated box (KRAB) at its N-terminal and a C2HC/C2H2 type zinc finger at its C terminals. This protein is highly conserved among human, mouse and rat, and widely expressed in many tissues.

Immunogen

synthetic peptide corresponding to an internal sequence of human ZNF746, conjugated to KLH. The corresponding sequence is identical in human ZNF746 isoform 2, and in mouse and rat ZNF746.

Application

Anti-ZNF746 antibody produced in rabbit has been used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

Zinc finger protein 746, also known as PARIS, parkin interacting substrate (ZNF746 ) accumulates in Parkinson′s disease (PD) models and in human PD brain. Hence it play a role in the progress of PD disease. ZNF746 is also regulated by the E3 ubiquitin ligase, parkin. In addition, overexpression of ZNF746 leads to the selective loss of dopaminergic neurons in the substantia nigra.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

A hallmark of Parkinson's disease (PD) is the preferential loss of substantia nigra dopamine neurons. Here, we identify a new parkin interacting substrate, PARIS (ZNF746), whose levels are regulated by the ubiquitin proteasome system via binding to and ubiquitination by
