General description

Ficoll-Paque PREMIUM products are a range of sterile, ready-to-use density gradient media for the preparation of mononuclear cells.

Ficoll-Paque PREMIUM products are based on Ficoll-Paque PLUS (see Ficoll-Paque PLUS) which has been used for more than 30 yr as a sterile density medium for the isolation of high yields of mononuclear cells from bone marrow, peripheral blood, and cord blood. All Ficoll-Paque PREMIUM products differ from Ficoll-Paque PLUS in that they are manufactured under a Quality Management System certified to ISO 13485:2003.

Application

Optimized for the isolation of mononuclear cells from human peripheral blood, bone marrow, and umbilical cord blood.

Can be used for isolating a broad range of human mononuclear cells including those of a higher density and for separating blood cells from mice or rats.

Can be used when isolating lower-density human mononuclear cells (e.g., mesenchymal stromal cells or monocytes).

Features and Benefits

  • Sterile, ready to use reagents used to prepare mononuclear cell preparations from peripheral blood, bone marrow, and umbilical cord blood by density gradient centrifugation.
  • Manufactured under a Quality Management System certified to ISO 13485:2003.
  • Low levels of endotoxin (< 0.12 EU/mL) secured and tested.
  • Cytotoxicity grade 0.
  • A Regulatory Support File (RSF) is available for Ficoll-Paque PREMIUM products.

The effects of the mononuclear cell separation method described by Bøyum procedure are noted below, since research situations may arise in which they are significant.

Protocols

The following procedure has been evaluated with Ficoll-Paque PLUS and is recommended for separation of normal blood samples for maximum reproducibility.

To prepare a Percoll gradient, the osmolality of Percoll must be adjusted with saline or cell culture medium to make Percoll isotonic with physiological salt solutions.
